Understanding Testobol

Testobol is a steroidal compound that contains 4,17beta-dihydroxy-4-androstene-3-one tetrahydropyranyl ether. Another name for this compound is 4-HydroxyTestosterone THP Ether (4-OHT Ether). 4-OHT is made in the body in very minute amounts from the metabolism of 4-hydroxy-4-androstene-3,17-dione (4-OHA) (Primobolan) and may also have a direct conversion from testosterone to 4-OHT. Since 4-OHA occurs in nature then its metabolites are also naturally occurring.

So now we know its a unique steroidal but what does it do and how should it be used? 4-OHT Ether is a steroidal that falls in into the 1-Androstenediol and 1-TESTOSTERONE category. It imparts a massive anabolic effect with a mild androgenic effect and therefore should be utilized with regular short cycle patterns (2 months on 1-2 months off).

Regular injectable testosterone and oral testosterone (Methyl Test) are considered an androgenic / anabolic hormones due to the fact that they possess a greater androgenic to anabolic ratio. 4-OHT Ether on the other hand is considered an anabolic / androgenic hormone due to the fact that it exerts more of an anabolic than androgenic effect. 4-OHT Ether is more anabolic and less androgenic than regular testosterone because of the addition of the 4-Hydroxyl group (4-OH).

THP Ether (Tetrahydropyranyl Ether) Etherification of steroids renders the compound fat soluble or lipophilic in order to allow absorption via the lymphatic circulation (same as long chain dietary fats). There can also be several different ether forms such as Tetrahydropyranyl, Tetrahydrofuranyl, Cyclopentyl, and Cyclohexyl etc.

Testobol is the first dietary supplement to contain actual Testosterone in the chemical name. Its a hydroxylated form of Testosterone that can be orally absorbed. Steroid hydroxylation occurs as a result of steroid biosynthesis via hydroxylases and cytochrome P450 enzymes. In other words steroid hydroxylation occurs during normal metabolism of androstenedione and testosterone. For instance 4-Androstenedione hydroxylation is known to occur at C6B, C11B, C15A, C16A, C16B, C19, and now at C4 and C3. (See example 1)

C4 hydroxylation allows the hormone to retain it properties while simultaneously increasing its anabolic activity and decreasing its androgenic activity. It also enables the compound to decrease estrogen and DHT biosynthesis from circulating testosterone and makes it chemically unlikely for the actual compound to directly convert into estrogen (See example 2 & 3). Another example regarding enhancement of anabolism would be the comparison of 4-HydroxyMethyltestosterone (4-OH MT) and Methyltestosterone (MT). According to U.S. Patent 3,060,201 by Camerino and associates, 4-OH MT has a myotrophic (anabolic) activity 3 times higher than MT while its androgenic activity is lower than that of MT.

Straight testosterone possesses extremely poor oral bioavailability. Androgens in general are neither truly water nor fat soluble compounds. This means if ingested orally almost none of the active compound enters the blood stream. When a straight androgen base is ingested orally it proceeds to the stomach (partial breakdown) and then to the intestinal mucosa where it is absorbed into the portal circulation and directly shuttled to the liver where it undergoes massive metabolic destruction.

Methyl Test or 17-alpha methyl testosterone is a 17-alpha alkylated form of Testosterone. This alkylation prevents the major route of androgen deactivation or oxidation to a 17-keto steroid. This allows a large portion of the active steroid to enter the bloodstream intact and avoid first pass liver deactivation, although this alkylation can also lead to hepatotoxicity.

Etherification on the other hand, appears to be a viable option to 17-alpha alkylation with no risk hepatotoxicity. The addition of various ethers enables the hormone to become fat soluble (lipophilic) so that it can be absorbed directly into the lymphatic system in order to bypass the destructive first pass of the liver. This is why it is important to take Testobol with a meal or fats. Long chain dietary fats are also absorbed via the lymphatic system and provide an enhanced transport of the active compound into the blood stream.


In summary, Testobol is the first supplement to contain an orally bioavailable form of real testosterone. This hydroxylated form of testosterone is more anabolic and less androgenic than real testosterone. Intro

Thats right, Promatrix Biotechnologies is proud to present THE FIRST LEGAL ORAL TESTOSTERONE SUPPLEMENT. Testobol is the second compound from the Hydroxy Hormone Series. Yes, I know thats real testosterone in the chemical name. How is this possible, you might ask? Well you see not to long ago we introduced Primobolan - 4-hydroxy-4-androstene-3,17-dione acetate (4-OHA) to the supplement world, which is considered by most pharmaceutical experts to be a drug. Actually, 4-OHA fits the criteria for dietary supplement like a glove. 4-OHA has never been sold in the U.S. as a drug and is found in nature. Now in order for us to receive FDA importation approval we needed to prove this so called natural occurrence. So we then demonstrated that C4(carbon 4) steroid hydroxylation occurs in nature and occurs with 4-androstene-3,17-dione (4-AD). 1 This represents a new era in nutraceutical hormones and the birth of several new patent pending dietary supplements.



Andro Metabolism
During the 4-OHA research I discovered that 4-OHA produced a C4-hydroxylated metabolite of Testosterone. Well I think you might know where I am going with this? Since, 4-AD naturally converts or produces 4-OHA then any metabolites of this compound would also be considered natural. Hence, the fact that humans and animals have been shown to produce 4-HydroxyTestosterone (4-OHT) from 4-OHA. Basically this means 4-HydroxyTestosterone can be sold as a dietary supplement.



Pharmaceutical Foresight
H. Levy in U.S. Patent 2,762,818 first discussed the synthetic manufacturing of 4-HydroxyTestosterone. 2 This patent was assigned to G.D. Searle & Co. in 1955 and consisted of eleven different methods for the synthesis of 4-OHT esters. It appears that they intended to use 4-OHT for its anabolic and androgenic properties. As with most other anabolic and androgenic steroids the medical use consisted of treating androgen deficiency and the promotion of nitrogen retention in metabolic processes. I have to say that Searle was definitely on the right track with this one. Although its a good thing they never brought this hormone to the drug market or we would still be searching for way to bring real testosterone to the supplement market.

4-OHT was one of the earlier attempts to make testosterone a safer drug (less androgen related side effects). Thus the C4-hydroxylation of certain C-19 androgens results in a dramatic decrease in estrogen and DHT conversion from the pro-drug while at the same time adding the extra benefit of inhibiting estrogen and DHT biosynthesis from plasma. Keep in mind that pro-steroid functionality always deals with the actual compound itself and the circulating parent hormone levels in this case testosterone levels. Lets use 4-AD as an example, now you must consider the conversion of 4-AD to estrone and testosterone but also consider the plasma testosterone conversion to estradiol and DHT.

C4 Steroid Hydroxylation
As you may already know 4-OHA (Primobolan) dramatically inhibits estrogen and DHT biosynthesis from circulating testosterone and cannot directly convert into estrogen while simultaneously stimulating natural testosterone production. On the other hand 4-AD directly increases estrone and slightly converts to testosterone but does nothing to inhibit estrogen biosynthesis from circulating testosterone and may even inhibit natural testosterone production if taken in a large enough dose. As I mentioned earlier steroid C4 hydroxylation of certain C18 and C19 androgens results in dramatic functional alterations of the parent hormone but even more importantly these hormones retain their anabolic / androgenic properties but with a shift toward greater anabolism rather than androgenicity.


For instance, 4-OHT does not directly convert into estrogen and it directly imparts anabolic / androgenic effects. It also has the ability to mildly inhibit estrogen and DHT biosynthesis from circulating testosterone. 4-OHT appears to be a more anabolic than androgenic version of real testosterone. So this means 4-OHT directly binds to the androgen receptor with a very high affinity and thus imparts its anabolic and androgenic effects. Another example of this would be the comparison of 4-HydroxyMethyltestosterone (4-OH MT) and Methyltestosterone (MT). According to U.S. Patent 3,060,201 by Camerino and associates, 4-OH MT has a myotrophic (anabolic) activity 3 times higher than MT while its androgenic activity is lower than that of MT. 3

Mild Estrogen and DHT Inhibition
So what we have here is a an highly anabolic / mildly androgenic version of orally bioavailable testosterone that also imparts a weak inhibition of estrogen and DHT from circulating testosterone. Marsh and associates studied the in vivo and in vitro metabolism of 4-OHA and its metabolites (4-OHT and 3-OH-dione) in rats. 4 3beta-hydroxyandrostane-4,17-dione (3-OH-dione) was identified as the major in vivo metabolite from 4-OHA while 4-OHT was the major in vitro metabolite. Researchers then tested the aromatization ability of all three compounds in order to determine if any of these metabolites might be contributing to 4-OHAs potent aromatase inhibitory activity. What they discovered was quite interesting. 4-OHA had an aromatase inhibitory activity of 91%, 4-OHT had an aromatase inhibitory activity of 59%, and 3-OH-dione had an aromatase inhibitory activity of 10%. This means 4-OHT exhibits only about 65% of the aromatase inhibitory effect of 4-OHA and that 3-OH-dione is too weak to even consider. This clearly demonstrates the superior aromatase activity of 4-OHA as compared to 4-OHT. The researchers concluded that the anti-aromatase activity of 4-OHA is largely due its own actions but its metabolites can not be excluded since 4-OHT exerts a weak aromatase activity.

Androgenic Potential

Androgenic potency was also evaluated utilizing the rat prostrate androgen receptor measured as the relative binding affinity (RBA). 4-OHA (RBA=.085) and metabolites A (RBA=.485) and B (RBA=.016) bound weakly to the androgen receptor while 4-OHT exhibited a significant relative binding affinity (RBA=75). 6 This RBA is comparable to potent androgens such as mibolerone (RBA=100) and 5-alpha dihydrotestosterone (DHT) (RBA=66). However, this assay did not distinguish between agonist and antagonist receptors. This lack of distinction inhibits the determination between the amount of androgenic and anti-androgenic effect. Although, 4-OHA research demonstrates that these receptors are somewhat agonist in nature meaning they impart an androgenic effect. It appears that 4-OHA has been shown to posses androgenic effects but these effect are attributed it 4-OHAs conversion into 4-OHT.
Ultra Extreme Anabolism
While there is limited research on 4-OHT there is enough to conclude that 4-OHT can impart potent anabolic and mild androgenic effects. For instance animal research performed by Gardi and associates tested the oral effectiveness of ether-modified hormones by measuring anabolic index and other variables. 4-HydroxyTestosterone cyclopentyl ether was orally administered in sesame oil to rats. The data collected for myogenic activity consisted of the anabolic index. Anabolic index is calculated as the ratio of increase in levator ani weight to the increase in the ventral prostrate weight. Ether modified 4-OHT exhibited a potent anabolic activity with the second highest anabolic index of (.78) out of 38 hormones tested. The control utilized to gage potency was the oral anabolic steroid methyltestosterone, which resulted in an anabolic index of (.37). This research demonstrates the ultra extreme anabolic potency of 4-Hydroxylated Testosterone ether. 7

4-OHT THP Ether

In order to enhance the oral bioavailability of our 4-OHT we decided to utilize the THP ether. Ether pro-steroids work on the premise of altering solubility. The chemical addition of the ether group renders the compound fat soluble or super lipophilic. Research demonstrates that ether technology is a viable variation to 17- alpha alkylation without hepatotoxic reactions or liver stress. The ether steroids are absorbed the same way most dietary fats (long chain fatty acids) are absorbed. After ingestion the body produces bile for emulsification and then lipase enzymes hydrolyze the compound to form what are called micelles. The micelles enter the intestinal cells where they form a lipoprotein carrier called chylomicron, which is a combination of ether steroid and special protein found in the small intestine. Once the compound forms the chylomicron it is released into the lymphatic system where it is then released into the blood stream. (A la intact steroid in the blood stream.) Remember the lymphatic system avoids the destructive first pass of the liver. A special note that most companies neglected to mention is that the research with ether steroids demonstrates oral administration with sesame oil. They mixed the active compound with sesame oil and then administered the compounds orally. The incorporation of the oil facilitated the lymphatic absorption since this is the same way most dietary fats are absorbed. 8 So basically this means you should take your Testobol with a meal or protein shake (just make sure to add a tablespoon of flax or olive oil to your protein shake).

Conclusion

Now you have been introduced to the first two new steroidals under the Hydroxy Hormone Series with the third compound (the most potent anabolic agent to ever enter the supplement world no kidding) due to arrive very soon. How do I gain weight in my upper body?

Actually I can answer this question quite simply because you just unknowingly started to answer it. First off take the body parts you mentioned in your question and train them accordingly: Monday - Biceps and Triceps, Tuesday - Chest and abs, Wednesday - Shoulders and traps. Perform 12 sets for biceps, 9 sets for triceps, 12-16 sets for chest, 8 - sets for abs, 12 sets for shoulders and 8 - sets for traps. Attempt 80-85% max weight with good form at 8-12 reps every set. Keep in mind you should train legs on Thursday. I say this only from experience because over time you might notice an unproportional development. But it is up to you. As far as your diet goes it might be a good idea to write every thing down that you put in your mouth for one week. Calculate the calories per day and then get a weekly average. This is the average amount calories you are consuming per day to maintain your current weight. When gaining lean mass you need to ensure more than adequate calories and protein for growth. 1 pound of muscle requires 2500 calories over your BMR vs. fat needing 3500 calories over your BMR. Add 500 calories per day to your daily average and try to consume 1 gram of protein per pound of body weight. This slight increase will not go to your waist because of your metabolism and training schedule.



Is creatine safe and how much is needed?

Yes. The average 200 lb. male bodybuilder can do quite well with just 5 grams (one teaspoon) post workout (after) with a good size meal. Actually 2-3 grams is all that is needed but the standard 5-gram dose is close enough. Within one-month muscle creatine concentrations should be reach maximum storage and thus you should notice increased weight, strength and anaerobic endurance.

If you stick with a top quality company like MVP Labs you are sure to get quality creatine. Another way to ensure quality is to look for a creatine manufacturer in the companys product literature. There are two top of line companies currently producing creatine. One is Pfanstiehl made in USA and the other is SKW Creapure made in Germany. If the company doesn't put in writing their raw material source then I wouldn't trust them. If they really were using one of the previously mentioned suppliers then it would behoove them to mention this in their advertising.

Actually it increases both muscle and water. A recent study measured muscle gain and the creatine subjects had significantly greater increases in Type I, Type IIA, Type IIAB muscle fiber cross-sectional areas. 1 Although this study didn't measure water as a variable other studies have concluded that creatine does increase water gain and that it is 90% intramuscular or inside the muscle cell. It's thought that this extra water may expand the muscle cell, increase mineral content, and turn on protein synthesis.

The only side effects are increased strength, muscle, water, and anaerobic endurance although an occasional trip to the toilet due to a 10-gram single dose has been noted, but we no better than that.

Creatine will not cause kidney damage or cramping. 2 A recent study revealed that creatine does not put athletes at risk for cramping no matter what the temp or humidity. They tested a large number of football players in hot humid conditions for several hours and no increased likelihood of cramping with the creatine group. I hope this helps. Which is better creatine liquid or powder? Is loading necessary? What about reloading? What do you think of HMB?

The main advantage of creatine powder over serum is that powder is stable and serum is not. I honestly do not know of too many truly stable forms of liquid creatine. A recently well known serum was tested by HPLC and was found to contain very little creatine and large amounts of creatinine. I hope you're not shocked at this but unfortunately this is what goes on in the supplement industry. No, creatine (powder) loading is not necessary. Just 2-3 grams post workout for 1 month will sufficiently load the muscles. Stopping creatine and reloading or restarting creatine supplementation is only necessary if you are concerned with natural creatine production. Research shows that continual creatine usage only slows down creatine production and does not shut down production. So whether you are coming off for health or performance reasons it really isn't necessary. But if you choose to come off then it would have to be at least 6-8 weeks for the creatine to completely leave the muscles. HMB has decent research showings it has the ability to increase strength and more recently decrease body fat. The muscle gains are another story. I haven't witnessed anyone develop noticeable muscle mass from HMB. But if you are combining it with creatine then I would give it a try.

I would give the HMB at least 1 month to produce results. But remember if you want to see if the HMB works for you then try it by itself without the creatine in order to establish if the HMB is producing the results and not some other supplement like the creatine.



Should I take a multi vitamin for more energy?

This is a very common question concerning energy levels. Although it does contains an internal flaw. You see micronutrients (i.e. vitamins and minerals) do not provide direct energy. The macronutrients (i.e. carbs, protein, and fat) on the other hand do provide a direct energy source. Although micronutrients do control the metabolic functions that control the metabolism of the energy nutrients (macro) thus providing energy. So if youre low or deficient in a particular vitamin then a multivitamin should help you to feel better. Another way to look at it is that vitamins help get the energy you need from food. It doesn't appear you need to increase your calories since you are with in IBW ideal body weight range.

The only other supplement that provides direct energy is herbal stimulants. I don't recommend this because you can become somewhat dependent upon them. Most companies that sell such compounds also contain an extensive health warning that should be read before purchasing such a product.

Try a one a day multi-vitamin multi- mineral like VITA-Plex from Promatrix Biotechnogies. If this doesn't help then go to your family physician for a routine check up.

Well this can be due to a couple of reasons. One it might just be that your total caloric intake is slightly to high and that your are genetically predisposed to accumulate fat in this area. Or you might be developing gynocomastia, which is usually associated with synthetic hormone use but has been documented in males that have never taken hormones. In these rare cases the individual usually experienced fat accumulation under the nipple, sore nipples, and slight swelling around the nipple during puberty and then had a recurrence later in life. If you have any of these symptoms then you might want visit your family physician for a routine checkup. Now if it is just excess body fat from increased calories then you need to adjust your macronutrient ratio by making sure your fat intake is between 20-30% and that your carbs are between 30-50% of total calories. Your total calories or BMR can roughly determined multiplying your body weight by 13. Keep taking your protein supplement but make sure that is either whey or casein and not soy.

Well it definitely sounds like your not eating enough. First you need to follow an appropriate macronutrient ratio: 60% Carb, 20% Protein, and 20% Fat. Next, we need to determine your total caloric intake. But first note that you really can not lose fat and not losing any weight. Understand that the reason you lost the weight and still have body fat stores is probably due to hypocaloric intake during intense aerobic exercise. This intense exercise predominately utilizes fat for energy but when carb and fat intake is insufficient the body will deplete glycogen stores and turn to protein for energy (muscle catabolism) thus leaving stores of fat while losing weight. If my metric conversions are right then you are about 10 pounds under IBW (Ideal Body Weight). Unless you wish to stay at your current weight I would recommend gaining weight (muscle) in order to be closer to your IBW.

Try to write every thing down that you put in your mouth for one week. Calculate the calories per day and then get a weekly average. This is the average amount calories you are consuming per day to maintain your current weight. When gaining lean mass you need to ensure more than adequate calories and protein for growth. Add 1000 calories per day to your daily average and try to consume .8 grams of protein per pound of body weight. This increase will not cause fat gain because of your intense training schedule. The increase in calories and carbs will allow for a protein sparing effect thus allowing your body to utilize your fat stores more efficiently for fuel.

The reason for the faster running time is probably due to the resistance training and being tired and losing weight is due to not enough total calories and carbs. These changes in your diet will not only improve your muscle mass and weight but it will also improve your athletic performance or run time. I am currently training to enter a full contact martial arts tournament have been following a bodybuilding program of one muscle a day but this makes me to sore to train and it is not defining me up as much as I would like to be. O.K. let me first say that sore muscles are good if you are trying to gain muscle size and strength. But of course if this is hampering your martial arts training then you need to look at whether this type of training is best suited for you and whether or not you are over training. As you might already be aware pre and post workout stretching of the target muscle can alleviate post workout muscle soreness. Also 200-400 IU of Vitamin E per day has also been shown decrease post workout muscle soreness.

If you are more concerned with muscle definition and endurance then there are some basic principals to abide by. Understand that it is probably not the one body per day routine causing the soreness but the sets, reps, and intensity that is usually associated with this type of program. For instance, your body parts can still be evenly divided throughout the week (i.e. Mon back - abs, Tues chest - calves, Wed biceps - triceps, Thur shoulders - traps, Fri quads - hamstrings) but your sets for large body parts should be between 12-16 and 9-12 for smaller body parts. Your reps should be between 15-30 and your weight should between 25-50% max weight. The tempo should fast at 30 sec - 1 min between sets. Now if I am totally off here then and you are already performing this type of routine then you should consider circuit training. This type of training is similar except that total sets are less and you are usually concentrating on upper body or even total body 2-3 times per week with the same fast light movements.

Cardio should be at least 3 times per week for 20 - 30 min. Depending upon your fitness level and past cardio training this can be increased slowly over time to 1 hour per day seven days a week. Also try to vary your intensity level every month or so by monitoring your max heart rate. One month try to stay at 75-80% then drop it for a couple of weeks to 55-60% so that your body does not adjust to the intensity level and thus curtails any future fat burning effects.


I have been weight training for several years with no improvements. How should I train in order to obtain my desired results?

From the info you have given me I am going to try to point you in the right direction. When you say desired results am I going to assume you mean increased size more so then definition. First lets make sure youre not over training. Try picking three mass exercises with three sets each for a total of nine sets. (Plenty for biceps, especially if your doing them twice a week) For instance take your current program and perform 3 sets barbell curls, 3 sets standing dumbbell curls, and 3 sets preacher curls. Drop the concentration (shape only) and incline aspect. Now keep the reps at 8-12 at 80% max weight with good form. Also make sure your biceps are not sore when you train the on the second day. If they are still sore on the second training day then you need to either drop the second training day or increase the biceps rest days. Next you need to make sure of the that all the aspects to growth are covered, which consist of calories, protein, and rest. Make sure your ingesting enough calories (body weight * 15) for growth, 1 gram of protein per lb. of body weight and no less then 8 hours of sleep. I hope this info will help in your quest for results. Give these tips at least 3 months to produce some type of response.

It demonstrates a 12 week cycle of Testo for 8 weeks and Primo starting at week 5 and Testo ending at week 8. Then just Primo is used for 4 weeks at which time you should discontinue use for at least 4-8 weeks. You should only use Testo and Primo simultaneously for 4 weeks out of the 12 week cycle.

Q: Is there the possibility of any kind of 'rebound affect' when use is stopped?

A: While this is a possibility I have not seen anything in the literature to directly or for that matter indirectly support this notion. Upon cessation of Primo the body will simply degradate the remaining active compound according to its half life which in turn allows the aromatase enzyme to regain normal estrogen production. Old patent research demonstrates that 4 weeks is the amount of time needed to regain normal estrogen production after cessation of Primo. This research did not mention any increase or lag in estrogen production which should have been noted by the researchers since they were monitoring estrogen levels in order to determine the amount of time needed to regain homeostasis.
